Alley, 12 Ocak 1951'de Wichita, Kansas'ta doğmuştur. 1976'da başladığı oyunculuk kariyerini 2022 yılındaki ölümüne kadar sürdürmüştür. 1987–1993 yılları arasında Rebecca Howe karakterini canlandırdığı televizyon dizisi Cheers ile tanınır. 1991 yılında bu dizideki rolüyle Emmy ödülü almıştır.
